Protein L affinity chromatography is typically used for the capture of antibody fragment drug substances (Fab, scFv, or sdAb containing a kappa light chain), since they generally lack affinity to Protein A ligand.
To meet regulatory requirements, detection and quantification of Protein L post-purification is a necessary step to ensure minimal protein L is present in the eluted drug substance. As leached Protein L is often in a complex with the antibody fragment, the samples must be subjected to a dissociation step to ensure accuracy of Protein L quantification.
